Western blot was performed using Anti-Desmin Polyclonal Antibody (Product # PA5-119736) and a 53 kDa band corresponding to Desmin was observed across the cell and tissue lysates tested except in MCF7, mouse kidney and mouse brain, and increased in expression in iPSCs upon differentiation to cardiomyocytes. Whole cell extracts (30 µg lysate) of RD (Lane 1), MCF7 (Lane 2), iPSC (Lane 3), iPSC differentiated to cardiomyocytes (Lane 4), and tissue extracts (30 µg lysate) of mouse skeletal muscle (Lane 5), mouse heart (Lane 6), mouse colon (Lane 7), mouse kidney (Lane 8), mouse brain (Lane 9) and rat skeletal muscle (Lane 10) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX), 12 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23002) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1500 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:10000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).
